고통을 느끼지 않는 아프리카 두더지쥐


     아프리카 두더지쥐는 많은 종류의 고통에 둔감하다. 약 10년 전, 독일 막스-델브뤽 분자의학센터(MDC) 연구진의 벌거숭이두더지쥐는 고통에 둔감한 이상한 감각 세계를 가지고 있는 것으로 유명해졌다. 연구자들은 이 아프리카 굴착 설치류가 통증에 현저하게 저항함을 증명했다. PLOS Biology  저널의 2008년 기사에 따르면, 그들은 두더지쥐가 칠레 고추에 있는 산이나 캡사이신에 노출되었을 때에도 고통을 느끼지 않는다고 보고했다. 이러한 특징은 아프리카 두더지쥐가 새로운 서식지에서 잘 적응할 수 있도록 한다.

As an international research team led by the MDC's Gary Lewin reports in Science, this characteristic has even allowed mole-rats to populate new habitats. Thanks to a genetic change, the highveld mole-rat is able to live alongside venomous ants with painful stings that other mole-rats avoid.


Around ten years ago, naked mole-rats at the Max Delbrück Center for Molecular Medicine (MDC) became famous almost overnight. That was thanks to the work of Gary Lewin from Berlin and Thomas Park from Chicago who together investigated the strange sensory world of the naked mole-rat. Park and Lewin showed that these African burrowing rodents were remarkably resistant to pain. In a 2008 article in the journal PLOS Biology, they reported that the mole-rats felt no pain when exposed to acid or capsaicin -- the substance that gives chili peppers their heat. The experiments on the mole-rats drew global attention.

남아공 프리토리아 대학의 연구팀과 Gary Lewin과 Thomas Park 연구진은 벌거숭이두더지쥐와 8종의 다른 관련 종을 대상으로 인간을 포함한 포유류의 피부에서 느껴지는 불타는 감각에 대한 실험을 시행했다. 연구팀은 세 개의 두더지쥐가 산에 둔감하다는 것을 발견했고, 이들 중 두 종은 캡사이신의 용액을 발에 주입했을 때 통증을 느끼지 않았다. 다른 하나는 발을 들거나 다리를 핥는 증상을 보였다. 또한 하나의 두더지 종이 아이소타이오사이안산알릴(AITC)에 영향을 받지 않는 것으로 나타났다. AITC는 와사비에 들어 있는 매운 성분이다. 이 종은 벌거숭이두더지쥐가 아니었지만 남아프리카공화국에서 발견된 다른 설치류인 하이펠트두더지쥐(Highveld mole-rat) 이었다. Lewin은 AITC는 신체의 아미노산을 공격하여 단백질을 파괴할 수 있기 때문에 이 발견은 매우 흥미로운 발견이라고 설명했다.

연구팀은 두더지의 통증에 대한 놀라운 저항성에 대해 분자적 이유를 밝히기 위해 실험된 9종의 동물의 척수와 지각 신경절로부터 감각 조직을 채취했다. 척수 신경절에는 통증 신호를 척수에 전달하는 뉴런 군집이 있다. 최첨단 시퀀싱 기술로 연구팀은 조직 내의 약 7,000개의 유전자 활성을 비교할 수 있었다. 


연구진은 통증을 느끼지 않는 동물 내에서 두 가지 유전자 활성이 변경되었음을 관찰했다. 통증 인식과 관련되어 있는 이온 채널 TRPA1과 NaV1.7 의 발현이 두더지쥐에서 크게 바뀌었음을 발견했다. 또한 연구원들은 AITC에 무감각한 종의 원인이 NALCN 채널 때문인 것을 발견했다. 채널을 차단하는 약물을 투여하여 NALCN 채널을 막았을 때 하이펠트두더지쥐가 갑자기 AITC에 민감하게 되었다고 설명했다. 


이 연구는 NALCN 채널의 높은 발현이 통증을 경감시키는 매우 효과적인 방법임을 입증하였으며, 새로운 진통제 개발 연구에 도움이 될 수 있을 것으로 보인다.
